Mitochondrial Phenotyping

Vi-Cell Automated Cell Viability Analyzer

The Vi-Cell S is a single-position cell viability analyzer manufactured by Beckman Coulter. The system automates the widely accepted trypan blue dye exclusion method for the determination of percent cell viability, cell number, size, and total cell concentration, greatly increasing the accuracy and reliability needed for identical cell plating in multiwell experiments. In addition to being fully automated, other unique features of this analyzer include the broad concentration range (5 x 104 to 1 x 107 cells/ml, a size detection range of 5 – 70 μm, a minimum sample volume of 1.0 ml, an analysis of the time of 3.5 minutes, and a 640 x 480 CCD array. The unit is also 21 CFR Part 11 enabled with self-contained, validated reagent packaging.

Anaerobic Chamber

It is a piece of lab equipment with a seal around it, allowing experiments to be performed in conditions without oxygen. The Type C Vinyl Anaerobic Chamber from Coy Laboratory Products is an environmental chamber/workstation suitable for conducting experiments under anaerobic (oxygen-free) conditions. This particular anaerobic chamber system has several unique features required for our use. The chamber is constructed of 20 mil. clear flexible PVC (polyvinyl chloride) vinyl, including optical quality, pressed polished 30 mil. PVC front panel for maximal visibility, and contains at least 26 cubic feet of interior volume (736 liters) and 8 square feet (.74 sq. meters) of workspace, including an airlock with a minimum of at least 1.4 cubic feet of pass-through space (39.6 liters). Unique functional features include a microcomputer-controlled, single pushbutton operation, front loading automatic airlock, with status lights and audible alarm for low gas. This airlock also includes backup independent manual controls which operate even when the automatic system is not functioning, another unique feature. The chamber airlock is also capable of the adjusting number of cycles (minimum of 81 different combinations of background and gas mix), enabling the user to adjust vacuum levels (10-30 in. Hg), display vacuum levels in either inches of mercury (Hg) or millibars, and to save a minimum of 9 different cycle configurations. Finally, this particular chamber system is equipped with a self-calibrating sensor and user-adjustable pressure adjustments for routine maintenance.

Seahorse XF24 Extracellular Analyzer

The XF24 Extracellular Flux Analyzer is a 24-well tissue culture microplate system that measures the oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of living cells simultaneously in real-time. Using new patented technology, the system provides completely non-invasive measurements without the addition of dyes, labels, or reporters. Released in late 2006, the XF24 Analyzer was and continues to be entirely unique to the biomedical research field.

The XF96 and XFe96 Extracellular Flux Analyzer

This machine is well suited for tissue culture microplate systems allowing for a larger number of assays. The systems can be used for screening and dose-response studies. The XF96 and XF e96 are validated for assays performed in hypoxia chambers and may be used with spheroid plates to study individual spheroids.
